MPxCaptureBeadswithintheMACSPlexCytokine12Kit,humanandMACSPlexCytokine10Kit,mousecontainacocktailofvariousfluorescentlylabeledbeadpopulations,eachcoatedwithaspecificantibodyreactingwithoneoftherespectivecytokineswithinthesample.
IncombinationwiththeExpressModesoftheMACSQuant®Analyzers(currentlyinstallExpressModes_26.1.12747)theMACSPlexCytokineKitsareoptimizedforautomatedmeasurementofbeads.Theysimplifyflowcytometricanalysisviapredefinedexperimentsettingsaswellasacquisitionandanalysistemplates.Theyapplyanautomatedgatingstrategytopopulationsofinterestthatwillbeautomaticallyadjustedforeachdatafileindividuallytoachieveoptimalresults.

MACSPlexCytokine12CaptureBeads,humanandMACSPlexCytokine10CaptureBeads,mouseconsistofbeadpopulationsthathavebeencoatedwithcaptureantibodiesspecificforcytokines.ThebeadpopulationscanbedistinguishedbydifferentfluorescenceintensitiesdetectedintheB1andB2channeloftheMACSQuant®AnalyzerandMACSQuantAnalyzer10(aswellasrespectivechannelsofcomparableflowcytometers).MACSPlexCytokineCaptureBeadsareaddedtounknownsamplesandtoserialdilutionsoftheMACSPlexCytokineStandard.Duringa2-hourincubationperiod,thecytokinesarecapturedbytheMACSPlexCaptureBeads.Subsequently,MACSPlexCytokineDetectionReagentcontainingamixtureofAPC-conjugatedanti-cytokineantibodies,isaddedinordertoformsandwichcomplexesduringa1-hourincubationperiod.Standardcurvesforeachofthecytokinesaregenerated.ThemedianoftheAPCfluorescenceofeachcapturebeadpopulationgivestheconcentrationofeachcytokineintheunknownsamples.
