Overview
CloneREA441recognizesthehumanandmouseIntegrinβ7antigen,asingle-passtypeImembraneproteinwhichisalsoknownasguthomingreceptorβsubunit.Integrinsareheterodimericglycoproteinreceptorsandexistasnon-covalentlyboundαandβsubunits.Theintegrin-β7subfamilyhastwoknownmembers(α4β7andαEβ7)thatareexpressedprimarilybyleukocytesandhavebeenimplicatedinsecondarylymphoidstructureformation,plasmacellshomingtogastrointestinalmucosa,andinflammatoryresponses.Theirligands:themucosalvascularaddressincelladhesionmolecule1(MAdCAM-1)andE-cadherinarepresentonthesurfaceofendothelialcells,BMstromalcells,andepithelialcells.Interactionofα4β7withMAdCAM-1allowsfortissue-specificmigrationofcirculatinglymphocytesintothelaminapropriaandPeyerpatchesofthegut,whereasαEβ7retainsintraepitheliallymphocyteswithinthegutepitheliumthroughE-cadherinbinding.Integrinβ7isalsoreportedtobeinvolvedinthepathogenesisofseveraldiseasessuchascolitis,diabeticinsulitis,andlymphoidmalignanciesincludinglymphomatouspolyposisinmantlecelllymphoma,thymiclymphoma,andmucosa-associatedTcellandBcellnon-Hodgkinlymphomas.
